The blue shifted plasmid samples from yesterday’s test was taken out of the red light exposure chamber and transformed with NEB turbo E.coli strain. Hopefully the full 24 hour exposure was enough to induce purine mutation in the naked DNA, we’ll find out soon.

Quick update on the day’s activity before the weekend. Growth comparison of the halomedia fungi contamination against YPD media yielded a somewhat disappointing result. The fungi seems to thrive in YPD with far slower growth rate in regular and acidic halomedia we have in stock.

I ran out of the original iteration of the kitchen media built around PPES-II using store bought milk and soy protein powder. The original media performed above expectations for the East River environmental sampling, but not quite so with the regular propagation of our in-house model organisms, notably brewer’s yeast and Deinococcus radiophilus.
